Do these nuts need thread lock in addition to the locking insert?

In most cases the nylon insert provides sufficient locking and you do not need thread lock. For extreme vibration or competition setups some users choose light thread lock on non-critical joints.

Can I reuse the nuts after removing wheels several times?

You can reuse them, but inspect the nylon insert after repeated removal. Replace any nut where the locking action feels reduced to avoid wheel loss.
